A 66-kDa protein of bovine hypophyseal Pars tuberalis induces luteinizing hormone release from rat Pars distalis Biocell
Lafarque,Martha; Oliveros,Liliana.
In this study, evidence for a factor secreted by bovine hypophyseal pars tuberalis that stimulates luteinizing hormone (LH) release from rat pars distalis cells is shown. The secretion products of bovine pars tuberalis cells into the culture medium were assayed on dispersed rat pars distalis cells in 30 min incubations and superfusion experiments. The culture medium from pars tuberalis total cell populations, added at a dose of 6 μg per tube, induced the greater LH release from pars distalis cells, without effect on follicle stimulating hormone (FSH) release. After pars tuberalis cells separation on a discontinuos Percoll gradient, only the culture medium of cells from 50 and 60% strength Percoll were able to release LH from rat pars distalis...

A cryopreservation protocol for immature zygotic embryos of species of Ilex (Aquifoliaceae) Biocell
Mroginski,Luis A.; Sansberro,Pedro A.; Scocchi,Adriana M.; Luna,Claudia; Rey,Hebe Y..
Tropical Ilex species have recalcitrant seeds. This work describes experiments demonstrating the feasibility of long-term conservation of Ilex brasiliensis, I. brevicuspis, I. dumosa, I. intergerrima, I. paraguariensis, I. pseudoboxus, I. taubertiana, and I. theezans through cryopreservation of zygotic rudimentary embryos at the heart developmental stage. The embryos were aseptically removed from the seeds and precultured (7 days) in the dark, at 27± 2ºC on solidified (0.8% agar) 1/4MS medium, [consisting of quarterstrength salts and vitamins of Murashige and Skoog (1962) medium] with 3% sucrose and 0.1 mg/l Zeatin. The embryos were then encapsulated in 3% calcium alginate beads and pretreated at 24 h intervals in liquid medium supplemented with...

A likely role for a novel PH-domain containing protein, PEPP2, in connecting membrane and cytoskeleton Biocell
Zou,Yi; Zhong,Wenping.
PH domains (pleckstrin homology) are well known to bind membrane phosphoinositides with different specificities and direct PH domain-containing proteins to discrete subcellular apartments with assistances of alternative binding partners. PH domain-containing proteins are found to be involved in a wide range of cellular events, including signalling, cytoskeleton rearrangement and vesicular trafficking. Here we showed that a novel PH domain-containing protein, PEPP2, displayed moderate phosphoinositide binding specificity. Full length PEPP2 associated with both plasma membrane and microtubules. The membrane-associated PEPP2 nucleated at cell-cell contacts and the leading edge of migrating cells. Overexpression of PEPP2 increased membrane microviscosity,...

A male-sterile mutation in soybean (Glycine max) affecting chromosome arrangement in metaphase plate and cytokinesis Biocell
Pires Bione,Nilton Cesar; Suely Pagliarini,Maria; Alves de Almeida,Leones.
A spontaneous male-sterile, female-fertile mutation affecting bivalent arrangement at the metaphase plate and cytokinesis was detected in line BR98-197 of the soybean breeding program developed by Embrapa - National Soybean Research Centre. Untill diakinesis, meiosis was normal with chromosome pairing as bivalents. From this phase, in several meiocytes, bivalents were not able to organize a single metaphase plate and remained scattered in the cytoplasm in a few or several groups. In these meiocytes, chromosomes segregated in both divisions giving rise to several micronuclei. However, the main cause of male sterility was the absence of cytokinesis after telophase II. Instead of the typical tetrads of microspores, four nucleate coenocytic microspores were...

A six-rhabdomere, open rhabdom arrangement in the eye of the chrysanthemum beetle Phytoecia rufiventris: some ecophysiological predictions based on eye anatomy Biocell
Meyer-Rochow,Victor Benno; Mishra,Monalisa.
We are describing a rhabdom organization of the eye of the chrysanthemum beetle Phytoecia rufiventris that to date has not been described from any other insect. In cerambycid beetles free rhabdomeres, forming a circular, open rhabdom, surround a central rhabdom made up of the rhabdomeres of one or two cells. In Phytoecia rufiventris the central rhabdomeres are missing throughout the eye and the microvilli of the outer 6 rhabdomeres are regularly oriented in three directions. Following the classification of rhabdom types suggested by Wachmann (1979), we suggest to name the rhabdom arrangement seen in the retina of Phytoecia rufiventris "Grundmuster 3". This pattern ought to facilitate polarization sensitivity and movement perception, features that agree...

A standard weight equation to assess the body condition of pejerrey Odontesthes bonariensis Biocell
Colautti,Darío C.; Remes Lenicov,Mauricio; Berasain,Gustavo E..
We developed a standard weight equation Ws to aid in the analysis of pejerrey Odonthestes bonariensis body condition over time and across populations using the relative weight index Wr. Weight - length data were compiled from 73 populations of pejerrey (N=16.022) from the Argentine pampas region. We used the regression-line-percentile technique, which provides a 75th-percentile standard by length intervals of 10 mm, to develop the Ws equation. The proposed equation is log10 Ws=-5,267+3,163 log10 Lst; Ws is weight in grams and Lst is standard length in millimeters. This equation is proposed for use with pejerrey between 120 and 520 mm of Lst. Values for Wr calculated with the Ws equation did not consistently increase or decrease as function of fish length,...

A study of chlorophyll-like and phycobilin pigments in the C endosymbiont of the apple- snail Pomacea canaliculata Biocell
Vega,Israel A.; Dellagnola,Federico A.; Hurst,Jorge A.; Godoy,Martín S.; Castro-Vazquez,Alfredo.
Pigments present in the brown-greenish C morph of an intracellular endosymbiont of Pomacea canaliculata were investigated. Acetone extracts of the endosymbiotic corpuscles showed an absorption spectrum similar to that of chlorophylls. Three fractions obtained from silica gel column chromatography of the acetone extracts (C I, C II and C III), were studied by positive ion fast atom bombardment-mass spectrometry (FAB-MS) and hydrogen-nuclear magnetic resonance (H-NMR). Results indicated the presence of (1) a sterol in the yellow colored C I fraction; (2) a mixture of pheophorbides a and b in the major green fraction, C II; and (3) a modified pheophorbide a in the smaller green fraction, C III. Aqueous extracts of the C endosymbiont did not show evidence of...

A tridimensional view of the organization of actin filaments in the central nervous system by use of fluorescent photooxidation Biocell
Capani,Francisco; Saraceno,Ezequiel; Boti,Valeria Romina; Aon-Bertolino,Laura; Fernández,Juan Carlos; Gato,Fernando; Krause,Maria Sol; Giraldez,Lisandro; Ellisman,Mark H.; Coirini,Héctor.
Cellular and subcellular organization and distribution of actin filaments have been studied with various techniques. The use of fluorescence photo-oxidation combined with phalloidin conjugates with eosin has allowed the examination of the precise cellular and subcellular location of F-actin. Correlative fluorescence light microscopy and transmission electron microscopy studies of F-actin distribution are facilitated with this method for morphological and physiological studies. Because phalloidin-eosin is smaller than other markers, this method allows the analysis of the three-dimensional location of F-actin with high-resolution light microscopy, three-d serial sections reconstructions, and electron tomography. The combination of selective staining and...

A unique mechanism of nuclear division in Giardia lamblia involves components of the ventral disk and the nuclear envelope Biocell
Solari,Alberto J.; Rahn,Monica I.; Saura,Alicia; Lujan,Hugo D..
The fine structure of the binucleate, parasitic protist Giardia lamblia during interphase and divisional stages was studied by serial thin sectioning and three-dimensional reconstructions. The earlier sign of nuclear division is the development of a few peripheral areas of densely packed chromatin directly attached to the inner nuclear envelope. An intracytoplasmic sheet of ventral disk components grows from the cell periphery towards one of the nuclei, apparently constricting this nucleus, which becomes located at a ventral bulge. After the basal bodies become duplicated, a full nuclear division occurs in trophozoites, giving two pairs of parent-daughter nuclei. This full division occurs in a dorsal-ventral direction, with the resulting nuclear pairs...

Age related interaction of dopamine and serotonin synthesis in striatal synaptosomes Biocell
Messripour,Manoochehr; Mesripour,Azadeh.
Tyrosine hydroxylase and tryptophan hydroxylase are key rate limiting enzymes in the biosyn-thesis of dopamine and serotonin, respectively. Since both enzymes are active in striatum, and affected by age, this study was undertaken to investigate interaction between dopamine and serotonin synthesis in brain striatal synaptosomes of aging rat. Male Wistar rats (3 and 30 month old) were killed by decapitation and brain striatal synaptosomes were prepared by discontinuous Ficoll/sucrose gradient technique. Synaptosomes were incubated in the presence of added pargiline (monoamineoxidase inhibitor), dopamine or serotonin synthesized during 25 min was measured by HPLC, employing electrochemical detection. Dopamine synthesis in synaptosomes prepared from young...

Agmatine inhibits hypoxia-induced TNF-alpha release from cultured retinal ganglion cells Biocell
Hong,Samin; Park,Kyoungsoo; Kim,Chan Yun; Seong,Gong Je.
The effect of hypoxia on the release of tumor necrosis factor-alpha (TNF-alpha) in transformed rat retinal ganglion cells (RGCs) and the effect of agmatine on the hypoxia-induced production of TNF-alpha in RGCs were evaluated. RGCs were cultured under hypoxic conditions with 5% oxygen, with or without 100μM agmatine. The expression levels of TNF-alpha and its receptor-1 (TNF-R1) were investigated by Western blot analysis. After 6 hours of hypoxia, we noted an increase in TNF-alpha production in RGCs. Agmatine significantly reduced TNF-alpha level after 12 hours of hypoxic treatment. The expression of TNF-R1 was not affected by the hypoxia or agmatine treatment. Our results show that agmatine inhibits the TNF-alpha production of RGCs in hypoxic...

Agmatine pretreatment protects retinal ganglion cells (RGC-5 cell line) from oxidative stress in vitro Biocell
Yoko,Iizuka; Hong,Samin; Yun Kim,Chan; Kab Kim,Seung; Je Seong,Gong.
Agmatine, 2-(4-aminobutyl)guanidine, has been reported to have neuroprotective effects against various neuronal damages. In this study it was investigated whether agmatine pretreatment rescues the retinal ganglion cells from oxidative injury in vitro. After differentiation of transformed rat retinal ganglion cells (RGC-5 cell line) with staurosporine, agmatine (0.0 to 100.0 μM) pretreatment was performed for 2 hours. Subsequently, they were exposed to hydrogen peroxide (0.0 to 2.5 mM) as an oxidative stress. Cell viability was monitored for up to 48 hours with the lactate dehydrogenase (LDH) assay and apoptosis was examined by the terminal deoxynucleotide transferase-mediated terminal uridine deoxynucleotidyl transferase nick end-labeling (TUNEL)...
